Fmoc-Gly-Wang Resin (100-200 mesh)

Fmoc-Gly-Wang Resin (100–200 Mesh): A Versatile Starting Point for Fmoc-SPPS

Fmoc-Gly-Wang Resin is a preloaded solid-phase synthesis resin, designed for Fmoc-based peptide synthesis, with glycine as the first amino acid already attached. Built on a polystyrene-divinylbenzene (PS-DVB) matrix and functionalized with a Wang linker, this resin is ideal for synthesizing peptides with a free carboxylic acid (-COOH) at the C-terminus.

The 100–200 mesh particle size offers an excellent compromise between mechanical stability and efficient reagent penetration, making it compatible with both manual and automated peptide synthesizers.

Product Specifications

  • Matrix: Polystyrene cross-linked with 1% divinylbenzene (DVB)

  • Functional Group: Wang linker preloaded with Fmoc-Glycine

  • Loading: Typically ~0.3–0.8 mmol/g (varies by supplier)

  • Particle Size: 100–200 mesh (~75–150 µm)

  • Swelling: Good swelling in DCM, DMF, and other aprotic solvents

  • Protecting Group: Fmoc (9-fluorenylmethoxycarbonyl), base-labile

Key Features

  • Preloaded with Fmoc-Gly: Eliminates the need for initial amino acid coupling, saving time and ensuring consistency.

  • Wang Linker: Acid-labile linker that releases peptides with a free C-terminal acid upon cleavage.

  • Fmoc Strategy Compatible: Fully suited to the widely used Fmoc/tBu peptide synthesis protocol.

  • High Purity and Consistency: Ensures reproducibility in peptide sequences and yields.

Applications

  • Peptide Synthesis: Ideal for synthesizing peptides starting with glycine at the N-terminus and terminating in a C-terminal free acid.

  • Structure-Activity Relationship (SAR) Studies: Streamlines the production of peptide analogs and libraries.

  • Bioconjugation Studies: Supports high-purity peptide synthesis for downstream labeling or conjugation.

Cleavage and Deprotection

  • Peptides can be cleaved from Fmoc-Gly-Wang resin using trifluoroacetic acid (TFA) in the presence of scavengers (e.g., water, TIS, EDT) to simultaneously remove protecting groups and release the free peptide.

  • Standard cleavage conditions: TFA cocktail (95% TFA, 2.5% water, 2.5% TIS) for 1–3 hours.

Storage and Handling

  • Store dry at room temperature, protected from light and moisture.

  • Swell in DCM, DMF, or other compatible solvents before use.

  • Avoid prolonged exposure to bases prior to synthesis to preserve the Fmoc group.
